Introduction:


Caffeic acid is widely distributed in various Chinese medicinal plants such as artichoke, cardoon, and honeysuckle. It is a phenolic acid compound with yellow crystal, soluble in hot water and ethanol.
It has effect on cardiovascular protection, anti-mutagenic, antibacterial and antiviral, lipid-lowering and hypoglycemic, anti-leukemia, and immune regulation. choleretic, hemostatic and antioxidant pharmacological effects.


Extraction Method


Caffeic acid is a common phenolic acid compound that has the effect of increasing white blood cells and is easily confused with caffeine. It is ubiquitous in the plant kingdom and is mainly derived from lemon peel, Ranunaceae plant Cimicifuga rhizome, valerian root and other plants. Caffeic acid, ferulic acid, sinapinic acid, and p-hydroxycinnamic acid are hydroxycinnamic acids that are almost universal in plants. These products have conjugated double bonds on the side chains of their molecular structures and are fluorescent under ultraviolet light. Most of them are blue. color fluorescence. This is a favorable condition for paper chromatography or thin layer chromatography examination.

Extraction method: Extract rhizomes of Cimicifuga single panicle with methanol, and concentrate under reduced pressure to remove methanol. Dissolve the residue by heating water, and filter out the insoluble matter while it is hot. After cooling, add benzene for extraction. The benzene solution is washed with 1% sodium bicarbonate aqueous solution, and the washing liquid is collected. Add dilute hydrochloric acid to acidify, use benzene and then free the organic acid from it, and concentrate under reduced pressure to remove benzene. The residue is a caffeic acid enriched product.


Preparation


To prepare crude caffeic acid
Take 30g of 3,4-dihydroxybenzaldehyde and 60.0g of malonic acid, add 300ml of N,N-dimethylformamide to dissolve, add 45mL of pyridine, and then raise the temperature to 70°C. Add 9mL of aniline and react for 1 hour. After the reaction is completed, cool the reaction solution to below 30°C, add 1000ml 2moL/L hydrochloric acid, stir, and stir at room temperature for crystallization for 2 hours. Filter with suction, and wash the filter cake twice with 300 ml of purified water. After blast drying at 50°C, 36.50g of crude yellow caffeic acid was obtained, with a yield of 93.60%, a purity of 99.52%, and a content of 99.2%.

Caffeine acid refining
Add 30g of crude caffeic acid and 120ml of ethanol into a 500mL four-neck bottle, raise the temperature to 80°C, stir until dissolved, add 1.5g of activated carbon for decolorization for 15min, and then filter while hot. Cool the filtrate to 5-10°C, stir and crystallize for 2 hours. Filter with suction, and wash the filter cake with 60 ml of purified water. After drying at 50°C, 27.39g of light yellow caffeic acid refined product was obtained, with a yield of 91.30%, a purity of 99.83%, and a content of 99.8%.
