Anti-obesity Effects and Homeostatic Regulation
A study investigated the anti-obesity effects and homeostatic regulatory role of 6-paradol in vivo. By comparing it with structurally similar vanilloid compounds-specifically 6-gingerol and 6-shogaol-researchers examined the physiological effects and the impact on genes related to fat metabolism in the adipose tissue and liver of mice with high-fat diet-induced obesity.
Sufficient quantities of the aforementioned vanilloid compounds were synthesized for animal experiments, involving oral administration to 6-week-old male mice over a two-week period. The results showed that 6-paradol reduced weight gain as well as visceral and subcutaneous fat accumulation in the mice within the two-week timeframe, whereas 6-gingerol and 6-shogaol did not produce these effects.
Furthermore, 6-paradol suppressed hepatic cholesterol and triglyceride levels and significantly downregulated the expression of genes associated with fatty acid synthesis, lipid transport, and adipocyte differentiation in both the liver and adipose tissue. Notably, while 6-gingerol significantly promoted the phosphorylation of AMP-activated protein kinase (AMPK)-a key regulator of lipid metabolism-6 paradol did not exhibit this effect.

Preparation Method
A method for preparing 6-Paradol comprises the following steps:
1. Preparation of the enone intermediate
Mix the raw materials-vanillin, 2-nonanone, and N,N-dimethylformamide-and react at 60°C–100°C. Monitor the reaction via liquid chromatography; stop the reaction when the content of the raw material 2-nonanone falls below 0.2%. Then, add an aqueous solution of glacial acetic acid, stir while cooling to room temperature, filter, wash with water, and dry to obtain the enone intermediate.
2. Preparation of 6-paradol
Charge a reaction flask with metallic sodium, anhydrous sodium acetate, and tetrahydrofuran. Under a nitrogen atmosphere, add the enone intermediate (dissolved in ethanol) to the flask. Upon completion of the reaction, decolorize with activated carbon, filter, and recover the solvent to obtain a pale yellow oily substance: 6-paradol.
