Description

Aloe-emodin is an anthraquinone compound with the chemical formula C15H10O5. It is mainly derived from the dried roots of the Polygonaceae plant Rheum palmatumL, Rheum tanguticum Maxim.ex Balf or Rheum officinale Baill. and rhizomes. Aloe-emodin is the active antibacterial component of rhubarb. Bacteria that are sensitive to it include Staphylococcus, Streptococcus and diphtheria, subtilis, anthrax, paratyphoid and dysentery, among which Staphylococcus and Streptococcus are the most sensitive. Aloe emodin also has the function of inhibiting the biosynthesis of DNA, RNA and protein in cancer cells.

Aloe-emodin

Physical properties:

Orange needle-like crystals (toluene), or earthy yellow crystalline powder. Melting point 223~224°, easy to sublimate. Soluble in acetaldehyde, benzene, hot ethanol, dilute ammonia, sodium carbonate and sodium hydroxide aqueous solution. Easily soluble in hot ethanol. It appears yellow in ether and benzene, and crimson in ammonia and sulfuric acid.

Preparation method: Aloe-emodin can be obtained by extracting the crude product from the low-temperature drying of aloe juice with ethyl acetate, and then purifying it through silica gel column chromatography (petroleum ether-ethyl acetate).

Antibacterial activity

At a concentration of 1.5 to 25 mg/ml, aloe emodin has inhibitory effects on Staphylococcus aureus, Streptococcus aureus, Bacillus diphtheriae, Bacillus subtilis, anthrax, Bacillus paratyphoid, and Shigella dysenteriae, among which Staphylococcus and Streptococcus are the most sensitive and antibacterial. The effective concentration is 15-25ug/ml. The in vitro inhibitory concentrations for Staphylococcus aureus 209P, Escherichia coli, and Shigella flexneri are 7.5 mg/L and 600 mg/L respectively. Their antibacterial effects are bacteriostatic rather than bactericidal. One of its mechanisms of action is to inhibit mitochondrial respiratory chain electron transport.

Aloe-emodin has a strong inhibitory effect on the nucleic acid and protein synthesis of Staphylococcus aureus. Aloe-emodin has a strong inhibitory effect on common clinical anaerobic bacteria. Among them, it can inhibit the growth of 90% to 100% of common Bacillus fragilis strains, and its MIC is slightly higher than that of metronidazole. At a concentration of 8 μg/ml, 76% to 91% of anaerobic bacteria can be inhibited.

Immunosuppressive effects

Aloe-emodin can inhibit the production of biological antibodies, inhibit the ability of carbon particles to clear away, reduce the weight of immune organs, reduce the number of white blood cells, and reduce the function of peritoneal macrophages. In vitro, it has a significant inhibitory effect on the incorporation of [3H]-TDR and [3H]Urd into lymphocytes at a concentration of 100 μg/ml.

Purgative effect

Aloe-emodin has strong purgative activity. Intestinal bacteria metabolize aloe-emodin, rhein, and rhein-anthrone, and the latter has strong purgative effect. Clinically used as a laxative, it has the effects of increasing appetite and relieving large intestine. According to foreign medical reports, aloe glycoside is hydrolyzed into aloe-emodin under the action of parasitic bacteria in the human body. This aloe-emodin stimulates the peristalsis of the intestinal wall. At the same time, due to the change in osmotic pressure, it is conducive to the elimination of waste in the intestine, thereby achieving irritation. Laxative, this stimulating purgative effect has special effects on constipation and hemorrhoids. Especially for middle-aged and elderly constipation, the therapeutic effect is more obvious.
