Methods
1) acid water extraction method
A) Weigh 10g of lotus leaf powder, add 200mLph2.5 hydrochloric acid solution in 80C water bath for constant temperature extraction for 2.5h, filter, filter the filter residue, then extract it with 100mL hydrochloric acid solution for 15h, filter, combine the filtrate twice, concentrate it to 50 mL by vacuum using a rotary evaporator, filter, adjust the pH value of the filtrate to 2~3, extract it with 60 mL chloroform in three times. Discard the chloroform layer to remove lipids and resins. Take the upper liquid, add 2%NaOH to adjust the pH to about 7, let it stand for a moment, wait for the precipitation visible to the naked eye, filter to remove the quality. The filtrate was then added with 2%NaOH to adjust the pH to 105, and then extracted with 90 mL of hydrogen form for 3 times. After evaporation and drying with chloroform liquid in a rotary evaporator, a small amount of pH25HCI solution was used to elute alkaloids on an ultrasonic cleaning machine, and then transferred to a 100ml volumetric bottle with a fixed volume, shaking to obtain the total alkaloid extract of lotus leaf and set aside.
B) Weigh 1g lotus leaf powder and put it into a round-bottom flask, add 35ml distilled water, adjust pH2 to 3 with 0.5% hydrochloric acid, connect a 40cm condensing tube to the round-bottom flask, and extract the acid liquid with 85C condensation reflux for 2h. After extraction, filter the extract with filter paper, extract the acid liquid with 30 ml chloroform in two times, and discard the chloroform layer. Add 0.5% sodium hydroxide to the acid water to adjust Ph7 and let it stand, filter it with filter paper, then add 05% sodium hydroxide to adjust pH9~11, then extract 60 ml chloroform in two times until there is no alkaloid in the water, combine the chloroform liquid, dry in the rotary evaporator, add 25 ml chloroform, and obtain by ultrasonic elidation
(2) Method: Weigh 5g lotus leaf powder, add 1g cellulase and 40mlcitric acid buffer solution with pH=5 at 50C.
