Ferulic acid contains reactive groups such as phenolic hydroxyl, carboxyl, olefinic bonds, and aromatic rings, making it a recognized natural antioxidant. Its physiological functions include free radical scavenging, antithrombosis, anti-atherosclerosis, antibacterial and anti-inflammatory effects, prevention of hypertension and heart disease, and enhancement of sperm motility. Therefore, it is widely used in medicine, food, and cosmetics.
However, due to its poor stability, ferulic acid is easily oxidized during storage and use, limiting the application and effectiveness of some of its important physiological functions. Therefore, in recent years, ferulic acid molecules have often been modified in some form to obtain more stable ferulic acid derivatives. Nyaradzo T. L. Chigorimbo-Murefu et al. studied the synthesis of esters from ferulic acid and natural compounds catalyzed by lipases. The results showed that *Candida albicans* lipase could perform transesterification reactions of ferulic acid vinyl derivatives and steroids, and that some of the synthesized compounds exhibited higher antioxidant activity than the precursor ferulic acid. Xin Jiaying, Zheng Yan, Zhao Guanli, and others first synthesized ethyl ferulic acid from ferulic acid and ethanol, and then reacted ethyl ferulic acid with vitamin E to obtain VE ferulic ester. VE ferulic ester is a compound formed by the esterification of ferulic acid with the 6-hydroxyl group of the β-chromone ring of α-tocopherol. Because it loses the free hydroxyl group, it does not possess the antioxidant activity of vitamin E and is relatively stable in air. It protects the 6-hydroxyl group of α-tocopherol, increasing the stability of α-tocopherol during storage and transportation. Simultaneously, it retains many physiological activities of ferulic acid, overcoming the disadvantage of its short alkane chain limiting its application in the oil and fat industry. Zhang Hui et al. modified ferulic acid with water-soluble chitosan, finding that chitosan enhances the activity of ferulic acid, and that FA-COS exhibits higher cytotoxic activity than ferulic acid.
The synthesis methods for ferulic ester derivatives include chemical synthesis, microbial transformation, and enzymatic biocatalysis. Enzymatic catalysis is considered a promising biotechnology due to its mild conditions, low byproduct rate, simple process, mild environment, high reaction selectivity, and easy product purification. Currently, ferulic acid glycoesters are mainly synthesized domestically through chemical methods. While chemical methods offer high conversion rates, they are complex, the products are difficult to separate, and produce numerous toxic byproducts, making continuous industrial production challenging. Research on enzymatic synthesis of ferulic acid glycoesters has not yet been reported.
