What is Buddleoside?
Buddleoside(linarin) is a natural chemical component widely found in medicinal plants such as wild chrysanthemum, buddleja, mint and buddleja. What does it do?
Antihypertensive effect
Chrysanthemum is used for a variety of diseases including hypertension. Its total flavonoids (luteolin and oleanolic acid) are believed to regulate vascular function and lower blood pressure. Studies have found that the combination of luteolin and oleanolic acid can inhibit the levels of angiotensin (Ang), aldosterone and endothelin, and increase serum NO concentration. After the combined administration of the two, systolic blood pressure (SBP) and diastolic blood pressure (DBP) decreased by 9.37% and 12.67%, respectively. In the in vitro vascular ring tension experiment, it was found that the vascular ring relaxation rate of oleanolic acid combined with luteolin (1:4) was higher than that of either alone, and the combination of the two could increase the expression of p-Akt and p-eNOS proteins, which may be through activating the PI3K/Akt/NO signaling pathway, enhancing the activity of eNOS, thereby increasing the synthesis and release of NO, and achieving the effect of vasodilation. Intragastric administration of 75 mg/kg linaloside can significantly shorten the dizziness time of spontaneously hypertensive rats and reduce SBP, DBP and mean blood pressure levels. It can also significantly inhibit the abnormal proliferation and migration of primary vascular smooth muscle cells VSMCs, and reduce the protein expression levels of AngII, angiotensin type 1 receptor (AT1), MMP-2, MMP-9 and p-Src, suggesting that linaloside may inhibit the activation of aortic AngII/AT1 signaling pathway, and then inhibit the abnormal proliferation and migration of vascular smooth muscle cells, thereby exerting an anti-hypertensive vascular remodeling effect.
Neuroprotective effect
Acetylcholinesterase (AchE) is an important target for the treatment of Alzheimer's disease (AD). Studies have found that linaloside has a selective
inhibitory effect on AchE in a dose-dependent manner. After KM mice were intraperitoneally injected with different doses of madagascar glycosides (35, 70 and 140 mg/kg), it was found that the AchE activity in the cerebral cortex and hippocampus of the mice was significantly reduced. The inhibitory effect of high-dose madagascar glycosides was equivalent to that of huperzine A treatment (0.5 mg/kg). Molecular docking studies showed that the 4'-methoxy and 7-O-sugar parts of madagascar glycosides play an important role in ligand-receptor binding and are mainly responsible for inhibiting AchE activity. Another study found that through molecular docking simulation, an aluminum chloride-induced zebrafish Alzheimer's disease model was established. Donepezil (DPZ) was used as a positive control drug to compare the effects of montanol on AD zebrafish movement disorders and AchE inhibition. The results showed that the movement disorder recovery rate and AchE inhibition rate in the 16.7 and 50μg/mL montanol (3d) groups were 88.0% and 74.5%, respectively, and the DPZ group was 79.3% and 43.6%. Therefore, montanol can not only significantly improve the recovery of movement disorders, but also significantly inhibit AchE activity. These research results provide certain experimental basis for the clinical application of montanol in the treatment of Alzheimer's disease.
Other effects
In addition to the above effects, 144μg/mL montanol also has antibacterial, analgesic, antipyretic, and anti-aging effects. The minimum inhibitory concentration of total flavonoids from wild chrysanthemum against Candida portugalensis is 61.6 μg/mL, while the minimum inhibitory concentration of lignoside is 18.0 μg/mL, with good in vitro antibacterial activity. Lignoside also has significant analgesic and antipyretic activity. Lignoside can significantly reduce the expression levels of senescence-related proteins p16, p21, and p53 in senescent cells, and the number of cells positive for the nuclear protein γ-H2AX protein associated with DNA damage is significantly reduced, indicating that lignoside can play an anti-aging role by eliminating the senescent phenotype of senescent rat bone marrow mesenchymal stem cells, and it is dose-dependent.
