A Fermentation Production Process
Currently, industrial fermentation production of succinic acid mainly employs an anaerobic metabolic pathway using microorganisms. The microbial cells first break down glucose into intermediates such as pyruvate through glycolysis, then fix CO₂ to produce oxaloacetate under the action of carboxylases. Subsequently, through metabolic steps involving malic acid and fumaric acid, succinic acid is reduced to succinic acid.
This metabolic pathway is highly dependent on CO₂, therefore the fermentation system typically requires a continuous supply of carbon sources and inorganic carbon. Traditional processes often use NaHCO₃, Na₂CO₃, or a continuous introduction of CO₂ combined with NaOH to adjust the pH to provide CO₂.
Currently, a fermentation process based on *Corynebacterium glutamicum* is attracting attention. This process typically begins with aerobic cultivation to rapidly enrich the microbial cells, followed by switching to anaerobic conditions for succinic acid fermentation. During the anaerobic stage, NH₄HCO₃, (NH₄)₂CO₃, or NH₄OH combined with CO₂ are used as the inorganic carbon supply, while simultaneously adjusting the pH. Compared to traditional sodium salt systems, this process can reduce the accumulation of sodium salt in the fermentation broth, reduce the pressure of subsequent waste liquid treatment, and reduce costs by utilizing a recyclable ammonium salt system, making it suitable for large-scale continuous production.
