What are Beta-alanine?

 

Beta-alanineBeta-alanine is a non-essential and naturally occurring amino acid. In nature, it exists in a free state in the root nodules of legumes, tea leaves, hydrolysates of mammalian brain tissue, and tobacco leaves and smoke. Beta-alanineBeta-alanine is a non-protein amino acid (not incorporated into protein during translation). It is synthesized in the liver and can be ingested through animal foods such as beef and chicken. Once ingested, beta-alanine combines with histidine within skeletal muscle and other organs to form carnosine, which functions to maintain acid-base balance in skeletal muscle, brain, and heart. Beta-alanine is the limiting factor in muscle carnosine synthesis.

 

Beta-alanine increases the concentration of carnosine in muscle. Carnosine scavenge reactive oxygen species and acts as an effective buffer to prevent the production of fatigue toxins in working muscles.

 

Reducing the production of fatigue toxins allows muscles to perform more repetitions, more sets, and more work before fatigue. Combined with a proper diet and exercise regimen, beta-alanine can therefore allow you to train harder, longer, and stimulate more muscle growth. Beta-alanine can also help improve performance in high-intensity cardiovascular events, such as sprinting and cycling.

 

Synthesis method


1. Acrylonitrile and ammonia react in diphenylamine and tert-butyl alcohol solution to generate β-aminopropionitrile, which is then subjected to alkaline hydrolysis. Add acrylonitrile, diphenylamine, and tert-butyl alcohol to a dry autoclave in sequence, stir for 5 minutes, add liquid ammonia, control the temperature at 100-109℃, the pressure at about 1MPa, and keep warm and stir for 4 hours. Cool to below 10℃, and stop stirring when the pressure drops to normal pressure. At 65-70℃/(8.0-14.7kPa), recover tert-butyl alcohol under reduced pressure to obtain crude β-aminopropionitrile, and then distill the crude product under reduced pressure to collect the 66-105℃/(1.33-4.0kPa) fraction to obtain β-aminopropionitrile. The alkaline hydrolysis operation is carried out in a reaction tank. First, add liquid alkali, control the temperature at 90-95℃-slowly add β-aminopropionitrile under stirring, and keep warm for 1 hour after addition. Evaporate under reduced pressure for half an hour to remove ammonia in the reaction solution, add appropriate amount of water, and drop hydrochloric acid to pH7-7.2. Filter to remove a small amount of insoluble impurities. The filtrate is concentrated under reduced pressure until a large amount of solid precipitates, and the material is discharged while hot, cooled to below 10°C, filtered, and vacuum dried to obtain β-alanine. This method consumes 982kg of β-aminopropionitrile per ton of product, and the yield of the alkaline hydrolysis stage is 90%.
2. Obtained by degradation of succinimide (Haugh reaction) Put alkaline sodium hypochlorite solution (containing 14% sodium hypochlorite, 8% sodium hydroxide, 30% sodium carbonate) and ice into the reaction tank, add succinimide under stirring, and react at 18-25°C for 0.5h. Heat to 40-50°C and react for 1h. Add hydrochloric acid to adjust to pH4-5 and concentrate under reduced pressure. After the concentrate is cooled, add 3 times the amount of 95% ethanol to precipitate inorganic salts, filter, and repeat once. Then dilute the filtrate with 4 times the amount of distilled water and reflux for 1h. Add activated carbon to decolorize, filter, and exchange the filtrate through exchange resin. Add activated carbon to decolorize the exchange solution, filter, concentrate the filtrate under reduced pressure, cool and crystallize, filter, and recrystallize once with distilled water to obtain β-aminopropionic acid.
3. β-aminopropionitrile method Obtained by hydrolysis and acid precipitation of β-aminopropionitrile.
4. It can be obtained by hydrolysis and purification of proteins such as sericin, gelatin, and corn slurry. It can also be synthesized by chemical methods.
5. Preparation method:
In a reaction bottle equipped with a stirrer, thermometer, and reflux condenser, add 1000g of 30% sodium hydroxide solution, heat to 90-95℃ while stirring, and slowly add 500g (7.14mol) of β-aminopropionitrile (2). After adding, continue to keep warm for 2h. Extract the generated ammonia under reduced pressure. Add an appropriate amount of water, adjust the pH to 7-7.2 with hydrochloric acid, and filter out the insoluble matter. The filtrate was concentrated under reduced pressure until solid precipitated, cooled to below 10°C, filtered, and dried in vacuo to obtain β-aminopropionic acid (1) with a yield of 90%.
