A study published in *Phytomedicine* (IF 6.7, Q1) systematically evaluated the mechanisms of action of licorice extracts and their active components in the context of colorectal cancer. Utilizing High-Performance Liquid Chromatography (HPLC) analysis, the study identified seven major constituents within licorice, among which Liquiritin was singled out as the key active substance. The analysis revealed that Liquiritin likely targets multiple tumor-associated proteins-including TP53, STAT3, SRC, and PIK3CA-and primarily modulates signaling pathways such as MAPK, PI3K-Akt, and apoptosis.

Through molecular docking and dynamics simulations, Liquiritin demonstrated a stable binding affinity for the TP53 protein, maintaining the structural integrity of the resulting complex via multiple hydrogen bonds. *In vitro* cellular experiments demonstrated that Liquiritin significantly inhibited the proliferation of SW480 colorectal cancer cells; concurrently, it suppressed their clonogenic potential, migration, and invasion capabilities, while also inducing apoptosis. Mechanistically, Liquiritin was found to downregulate the expression of invasion-associated proteins, such as MMP-1, MMP-3, and MMP-9.
In *in vivo* animal studies, Liquiritin treatment significantly inhibited the growth of xenograft tumors in nude mice, evidenced by a marked reduction in both tumor volume and weight. Furthermore, histological examination of tumor tissues revealed structural disruption and areas of necrosis; notably, no significant changes in body weight or signs of toxicity were observed. The study suggests that Liquiritin exerts its effects by upregulating p53 activity and inhibiting the phosphorylation levels of the p38 MAPK signaling pathway, thereby modulating the proliferation and survival of tumor cells.
A comprehensive analysis of these findings suggests that Liquiritin likely exerts its multi-targeted anti-tumor effects by modulating the p53/p38 MAPK signaling axis. This provides a robust scientific foundation for the potential application of Liquiritin in the development of functional ingredients and plant-derived active therapeutics.
