Mechanism of Action and Data

 

 

1. Inhibition of Melanin Production

Ascorbyl Tetraisopalmitate inhibits melanin production by suppressing tyrosinase activity. Human melanoma cells (HM-3-KO) were cultured with VC-IP at concentrations ranging from 0% to 0.2% for 4 days. Cells were then collected using trypsin and centrifugation. After cell counting, melanin density was measured using a Pharmacia laser densitometer. Compared to the control group, melanin synthesis in the VC-IP group decreased from 72% to 12%.

2. Inhibition of Matrix Metalloproteinase (MMps) Activity

VC-IP significantly inhibited MMP activity and suppressed UV-induced collagenase elevation. NHDF cells cultured in serum-free medium at 50 μm for 48 hours were ultrafiltered, concentrated, and cultured on an SDS-polyacrylamide gel containing 0.2% gelatin under non-reducing conditions. The gels were then stained with Coomasie Brilliant Blue R250 and measured using a laser densitometer. When using VC-IP (20-50µm), the activities of MMP-2 and MMP-9, which degrade gelatin, were inhibited to 18%-23% and 10%-17%, respectively. In contrast, the MMP activities in the VC (20-50µm) group were 39%-46% and 42%-52%, respectively.

3. Promotion of Collagen Synthesis

UV-induced release of collagenase from infiltrating cells leads to accelerated enzymatic degradation. In this case, the rate of collagen degradation exceeds the rate of biosynthesis, resulting in a reduction in collagen. Human skin fibroblasts (NHDF) cells were treated with L-[2,3H]proline for 24 hours, followed by cell lysis and subsequent neutralization with NaOH. The obtained cell extracts were thoroughly hydrolyzed with *Clostridium perfringens* collagenase, treated with TCAtannin, and centrifuged to produce a supernatant. The precipitate consisted of collagen and non-collagen components, which were evaluated using an Aloka liquid scintillation counter LC-3600 sci. VC-IP increased collagen synthesis in human skin fibroblasts by 170%-195% within a concentration range of 20-50 μm, while VC increased collagen synthesis by 112%-127% within the same concentration range.

4. Thermal Stability Test

VC-IP was stored at a constant temperature of 40℃ or 60℃ for one month. No significant changes were observed in appearance or color. The weight of VC-IP measured at 200℃ and heated at a rate of 5℃/min for temperatures ranging from 30-500℃ remained unchanged. These results indicate that VC-IP possesses good thermal stability.
