What is gamma cyclodextrin?

 

Gamma Cyclodextrin (γ-cyclodextrin, γ-CD) is formed by cyclodextrin glycosyltransferase acting on glucose polymers such as starch, glycogen, malt, and oligosaccharides. It is a cyclic oligosaccharide composed of 8 D-pyranose glucosides connected head to tail by α-1,4-glycosidic bonds. It has a unique barrel structure with external hydrophilicity and internal hydrophobicity, and can be used for solubilization and stabilization of guest molecules. At present, γ-CD is mostly used as a raw material for the production of sodium sugammadex. Sodium sugammadex is often used as an injection for neuromuscular blocking muscle relaxants, which can cause allergic reactions and hypersensitivity reactions in patients, and the allergic reactions are induced by large molecular exogenous proteins. γ-cyclodextrin is mainly obtained by enzymatic hydrolysis of starch by cyclodextrin glycosyltransferase (cyclodextrin glycosyltransferase, CGT enzyme, EC2.4.1.19).
γ-cyclodextrin is a polymer chemical product. γ-cyclodextrin has a larger cavity than β-cyclodextrin, so the cavity can contain a wider range of guest molecules; it has better water solubility. At room temperature 25°C, the solubility of γ-cyclodextrin is 25.6g/100ml, while the solubility of α-cyclodextrin is 12.7g/100ml water and the solubility of β-cyclodextrin is only 1.88g/100ml water. It has a broader application prospect than β-cyclodextrin.

 

Main uses

 

1. Uses in the pharmaceutical industry
Cyclodextrin can be used to form inclusion compounds (encapsulation) with drugs

(1) Stabilization of unstable drugs;

(2) Powderization of deliquescent, sticky or liquid drugs;

(3) Solubilization (solubilization) of poorly soluble or insoluble drugs, etc.

2. Uses in the pesticide industry
Cyclodextrin inclusion stabilization can make some pesticides durable in storage and improve their insecticide efficacy.

3. Uses in the food industry
Cyclodextrin is used in the food industry to have the following effects:

Cyclodextrin is a valuable chemical reagent that can be used as a carrier for flavors, sweeteners and pigments. It is loosely encapsulated, highly soluble, and can be easily decomposed into glucose, so it is particularly suitable for food.

 

Gamma Cyclodextrin has a cavity diameter that can encapsulate many substances that α-CD and β-CD cannot encapsulate; γ-CD has the highest solubility and can most effectively improve the solubility and emulsification ability of the guest compound; in addition, γ-CD is the safest for the human body and can also be acted upon by enzymes, making it easily degraded and absorbed by the small intestine, thereby reducing renal toxicity.

Based on the above advantages, the application points of γ-CD can be summarized into four aspects:
(1) Masking bad odors;
(2) Improving bioavailability;
(3) Controlling the release rate of guest molecules;
(4) Improving product solubility and stability. Coenzyme Q10 plays an important role in mitochondrial electron transport and has been used to treat heart disease and degenerative diseases. However, coenzyme Q10 is a fat-soluble compound that cannot be well absorbed by the human body after oral administration. Therefore, its solubility and bioavailability are improved by encapsulating it in γ-CD, making it a nutrient for anti-aging and improving health. Curcumin is a yellow polyphenol isolated from the rhizome of turmeric, which has shown therapeutic effects in treating cardiovascular diseases, diabetes and Alzheimer's disease. However, the oral bioavailability of curcumin is less than 1%. After being encapsulated in γ-CD, its bioavailability reaches 85% or more.
