What is Caffeic Acid Phenethyl Ester?

 

Caffeic acid phenethyl ester (CAPE) is one of the main active components of propolis, possessing antiviral and antioxidant properties. It has inhibitory effects on herpes viruses, adenoviruses, and influenza viruses. Various compounds in propolis, including CAPE, quercetin, isopentenyl esters, rhamnetin, galangin, glycosides, and polysaccharides, exhibit anticancer activity.

The structure of CAPE contains an o-dihydroxy (catechol) phenyl group, a common free radical-scavenging structure. The presence of two adjacent phenolic hydroxyl groups increases the electron density of the benzene ring, making it prone to auto-oxidation and coloration. The chemical composition of propolis varies by region, and the CAPE content differs accordingly. Chinese propolis has a relatively high CAPE content (15–25 mg/g), whereas Brazilian propolis does not contain CAPE.

In the laboratory, CAPE can be synthesized using caffeic acid and β-bromoethylbenzene as raw materials, with potassium phosphate as a catalyst, achieving a yield of 60%.

Chemical Structure of CAPE

Caffeic acid phenethyl ester (CAPE) contains an ortho-dihydroxy (catechol) phenyl group, a common structural feature for free radical scavenging. The two adjacent hydroxyl groups in this structure increase the electron density of the benzene ring, making it prone to auto-oxidation and coloration.

According to research by Bors et al., the ortho-dihydroxy benzene structure plays a crucial role in the antioxidant activity of flavonoid compounds. Additionally, the double bond at the 2',3' positions in the CAPE molecule is relatively reactive, further enhancing its antioxidant properties.

The phenethyl ester group in CAPE is also essential for its biological activity, as it improves CAPE's ability to cross cell membranes, thereby facilitating its physiological and pharmacological effects.

Antioxidant Activity
Caffeic acid phenethyl ester (CAPE) exhibits strong antioxidant activity due to the presence of its catechol group. Studies have shown that CAPE can completely block the production of reactive oxygen species (ROS) in neutrophils at low concentrations (10 μM) and inhibit the xanthine oxidase (XO) system.

The main antioxidant components in propolis include CAPE, caffeic acid, tannins, kaempferol, galangin, and cinnamyl caffeate. Propolis containing CAPE exhibits stronger antioxidant activity, and CAPE has been found to have superior antioxidant capacity compared to galangin.

According to Ozyurt et al., the antioxidant effects of propolis extracts are stronger than vitamin E, with CAPE being one of the key active components. CAPE exerts its antioxidant effects by scavenging ROS, inhibiting XO and nitric oxide synthase (NOS), and reducing the consumption of superoxide dismutase (SOD).

Immunomodulatory and Anti-Inflammatory Effects
CAPE exhibits significant anti-inflammatory properties and has been shown to be effective in both acute and chronic inflammation models, such as formalin-induced arthritis and carrageenan-induced edema.

Additionally, CAPE plays an immunomodulatory role by affecting immune organs like the thymus and spleen, reducing thymus weight, and decreasing T-cell count.

Cardiovascular Regulation
CAPE influences vascular function through a dual mechanism:

  • At low concentrations, it stimulates nitric oxide (NO) release, promoting vasodilation.
  • At high concentrations, it inhibits intracellular and extracellular calcium ion exchange, thereby regulating vascular function.

These effects suggest that CAPE has potential applications in the prevention of cardiovascular diseases and post-surgical complications.
