Reishi Extract Powder: Evidence for delaying vascular aging
A study by a research team from Hainan Medical University has found that ganoderic acid D (GA-D), an active ingredient in Ganoderma lucidum, can restore lysosomal function by stabilizing the VAPB-SNX25 linker complex, thereby delaying vascular aging.
The researchers first discovered that GA-D significantly improved cell viability in a D-galactose-induced vascular smooth muscle cell (VSMC) aging model and reduced the expression of p16, p21, SA-β-gal, and reactive oxygen species. Flow cytometry analysis showed that most D-galactose-treated VSMCs were arrested in the G0/G1 phase, while the proportion of cells in the S or G2/M phase decreased, and GA-D attenuated this phenomenon. These results suggest that GA-D has the potential to alleviate VSMC aging.
The researchers then evaluated the effects of GA-D on vascular function in aged mice. The results showed that the pulse wv. value (PWV) was significantly increased in aged mice compared to younger mice. This age-related increase was attenuated in a dose-dependent manner by GA-D treatment. Similarly, GA-D intervention reduced SA-β-gal, aging markers, and reactive oxygen species levels in senescent blood vessels and significantly improved the structure and function of senescent blood vessels.
GA-D improves lysosomal function by enhancing the interaction between VAPB and SNX25 in senescent vascular smooth muscle cells, thereby promoting endoplasmic reticulum-mediated lysosomal membrane repair. Enhanced lysosomal function in senescent cells further promotes autophagy, improving vascular remodeling and function; VAPB is a key factor in maintaining VSMC lysosomal homeostasis.
In summary, this study reveals the precise regulatory role of GA-D in improving vascular aging, suggesting that GA-D holds promise as a candidate drug for clinical intervention in vascular aging. While GA-D can alleviate age-related vascular functional decline, its long-term impact on overall lifespan still needs to be evaluated in future studies.
