Protocatechuic acid (PCA) is an organic compound with a molecular formula of (HO)2C6H3COOH and a molecular weight of 154.12. It is a white to brown crystalline powder that changes color in the air. It is soluble in 50 parts of water; soluble in ethanol and ether. It exists in the leaves of Stenoloma chusanum (L.) Ching, a plant of the Lepidoptera family, and the leaves of Ilex chi-nensis Sims, a plant of the Aquifoliaceae family.

Protocatechuic acid is extracted and separated from Phellinus lonicerinus, which was collected in Yichang, Hubei Province and identified as Phellinus lonicerinus (Bond.) Bond.et sing by Professor Mao Xiaolan of the Institute of Microbiology, Chinese Academy of Sciences, 95% industrial alcohol.

The dried fruiting bodies of Phellinus lonicerinus were crushed, and 2 kg of Phellinus lonicerinus powder was extracted by ultrasonic assisted extraction with 10 liters of 95% ethanol. The extraction was performed three times, each for three hours, and the extracts were combined and concentrated under reduced pressure at 45°C to obtain the alcohol extract of Phellinus lonicerinus. The extract was suspended with water, and polyphenols were enriched on macroporous adsorption resin, and gradient eluted with 95% ethanol (0, 20%, 40%, 60%, 70%, 80%, 95%). The eluate was qualitatively identified for polyphenols using ferric chloride colorimetric agent, and the fractions that may contain polyphenols were combined, concentrated to dryness at 45°C using a rotary evaporator, and weighed to obtain total phenols from Lonicera japonica, wherein 2 kg of Lonicera japonica produced about 8 g of total phenols, with an extraction rate of about 4‰.
