Promoting skin regeneration

The researchers applied 0.5% hexapeptide-9 and vitamin C to human skin or human fibroblast cultures in vitro, and treated other human fibroblast cultures with 0.75% hexapeptide-9 for 16 hours. Immunostaining was used to stain the components such as filaggrin, integrin β1, type IV collagen, laminin, and type I collagen, and then observed using an electron microscope. Among them, green immunostaining can observe the thickness of skin proteins after treatment in each experimental group; blue or red fluorescence indicates cell nuclei.

Experimental data showed that at the dermal level, hexapeptide-9 can quickly trigger an increase in the expression of type I and type III collagen (data not shown), and promote the synthesis of type IV collagen and laminin 5. In addition, improvements in some epidermal differentiation markers (filaggrin, integrin) were observed.

Promoting tissue repair

After treating damaged skin with 0.5% hexapeptide-9 for 72 hours, the researchers compared it with untreated skin. Under the microscope, it can be seen that the damaged cracks are repaired obviously, indicating that hexapeptide-9 can promote rapid and complete regeneration of the epidermis and has a good repair effect on the skin.

Human trials

The researchers recruited 20 healthy female volunteers aged 20-62 to conduct clinical trials: a gel containing 1.5% hexapeptide-9 (1% GP4G IS) was applied to the crow's feet twice a day for 4 weeks. The skin wrinkles were quantitatively analyzed and compared with the placebo gel. The results showed that compared with the placebo group, the hexapeptide-9 group had improvements in the total skin surface, number of wrinkles, total length of wrinkles, and average depth of wrinkles, proving that hexapeptide-9 can reduce the number and depth of wrinkles.
