Procyanidin B2, chemically known as epicatechin-(4β-8)-epicatechin, is a type of proanthocyanidin (PC) widely found in various plants such as grape seeds, peanuts, apples, hawthorn, and tea.It exhibits high biological activity and stronger antioxidant capacity than vitamins C and E. Studies have shown that procyanidin B2 plays an important role in anti-inflammation, regulating blood lipids and blood sugar, and protecting the cardiovascular and nervous systems. Furthermore, procyanidin B2 (PB2) is also one of the most representative natural food colorings.

PB2 is widely found in the flowers, fruits, leaves, and seeds of various edible plants, including those from the Rosaceae, Fabaceae, Nepeta, and Rhamnaceae families. Fruits of the Rosaceae family, such as hawthorn (Crataegus pinnatifida Bge.), apple (Malus pumila Mill.), European pear (Pyrus communis L.), peach (Prunus persica (L.) Batsch), and mugua (Chaenomeles Lindl.), are nutritious foods in people's daily diets and are also important sources of PB2. The concentration of PB2 in hawthorn fruit is as high as 243 mg/100g dry weight (DW), far exceeding that of catechins, coumaric acid, hyperoside, and isoquercitrin. Furthermore, PB2 has been found to be the second highest polyphenol content in traditional apple peel, at 954 ± 343 μg/g DW, and the highest among commercial apple varieties.

Proanthocyanidins B2 possess numerous biological activities, including anti-inflammatory effects, anti-obesity and anti-diabetic effects, anti-diabetic complications effects, liver protection, neuroprotection, gastrointestinal protection, vascular protection, kidney protection, and reproductive system protection. It also has the potential to treat/prevent various human diseases, such as diabetes, diabetic complications, atherosclerosis, and non-alcoholic fatty liver disease.

Antioxidant Effects

Proanthocyanidins B2 has extremely strong antioxidant capacity, effectively scavenging reactive oxygen species such as superoxide anion radicals and hydroxyl radicals, thereby delaying aging and preventing various chronic diseases.

Proanthocyanidin B2: Effects on androgenetic alopecia (AGA)

 

Researchers evaluated the efficacy and safety of topical proanthocyanidin B2 in treating male alopecia areata (AGA).

 

The study included 40 eligible male patients aged 18 to 50 years with AGA and Hamilton-Norwood classification II-V. Patients were randomly assigned to receive either topical proanthocyanidin B2 (n=20) or placebo (n=20) for 16 weeks. After treatment, patients returned for follow-up evaluation. The efficacy of topical proanthocyanidin B2 was evaluated using follicle microscopy and whole-body photography.

Results

 

Forty male patients with AGA II-IV (mean age 33.32 years, range 21-44 years) were included. There were no significant differences between the two groups in terms of age, duration of alopecia, and AGA classification (P > 0.05). At the end of the study, the topical proanthocyanidin B2 group showed a significant increase in total hair count compared to baseline compared to the placebo group (P < 0.05). Hair count during the growth phase was also significantly increased in the proanthocyanidin B2 group (P < 0.05).

 

This placebo-controlled study suggests that topical proanthocyanidin B2 is an effective and safe treatment option for patients with AGA.
