Experimental Procedures

Researchers extracted the effective components of black cumin seeds using the ethanol maceration method, and obtained lyophilized black cumin seed extract powder after concentration and freeze-drying. In vitro experiments were conducted using mouse B16F10 melanoma cells, and an oxidative stress model was established with 0.4 mmol/L H₂O₂. Cell viability, tyrosinase activity, melanin content, SA-β-gal senescence staining, scratch healing ability, ROS level, mitochondrial membrane potential, antioxidant enzyme activity, and the expression of proteins related to signaling pathways such as MAPK, PKA, Wnt/β-catenin, and Nrf2-ARE were measured. In vivo experiments used a PTU-induced zebrafish pigment inhibition model to observe the effect of black cumin seed extract on promoting melanin deposition in vivo. In addition, the experiment also evaluated the effects of black cumin seed extract on cellular senescence marker proteins p53/p21, dendritic structure (F-actin staining), cell migration-related proteins (Rac1/2/3, Cdc42), and cell adhesion protein (E-cadherin).

Experimental Conclusions

The results showed that black cumin seed extract significantly increased tyrosinase activity by approximately 1.5 to 2 times, increased melanin content by 30% to 50%, and upregulated the expression of key melanin-producing proteins such as Tyr, Mitf, and gp100. Zebrafish experiments also confirmed that black cumin seed extract can effectively reverse PTU-induced pigmentation inhibition and restore melanin deposition on the skin surface.

Mechanistically, Black Seed Extract synergistically promotes melanin synthesis by rapidly activating the MAPK pathway (p38, ERK1/2, and JNK phosphorylation peaks at 5 minutes), increasing p-CREB levels in the PKA pathway, and inducing Wnt/β-catenin pathway activation. Meanwhile, black cumin seed extract exhibits significant antioxidant effects, reducing ROS levels by 40% to 60%, restoring mitochondrial membrane potential, increasing the activity of antioxidant enzymes such as SOD, CAT, and GSH-Px, and reducing MDA content by up to 35%. Its mechanism is closely related to the activation of the Nrf2-ARE pathway, promotion of Nrf2 nuclear translocation, and upregulation of HO-1 and NQO1 expression.

Furthermore, black cumin seed extract can effectively inhibit oxidative stress-induced premature cell aging, reducing the proportion of SA-β-gal positive cells from 45% to 20%, downregulating p53/p21 expression, improving cell migration ability (scratch healing rate increased by 2-fold), promoting dendritic structure repair (length increased by 50%, number returned to normal), and increasing E-cadherin expression by 2.5-fold, enhancing cell matrix adhesion.

In summary, black seed extract, through a mechanism of "promoting melanin synthesis + anti-oxidation and anti-aging," repairs melanocyte function from multiple dimensions, providing a scientific basis for the development of vitiligo treatments.
